7/ These findings highlight new strategies to combat metastatic recurrence, targeting both epigenetic regulation and immune interactions. Restoring Brd7 expression or modulating the immune response could be the future of metastasis treatment.
6/ The good news? This process can be reversed! Inhibiting NET formation with sivelestat or depleting neutrophils significantly reduced metastatic growth. Blocking CTLA-4 (an immune checkpoint) also restored anti-tumor immunity.
5/ Neutrophils aren’t just bystanders. In this scenario, they create an immunosuppressive niche by releasing neutrophil extracellular traps (NETs), which help metastatic cells survive and contribute to T cell exhaustion.
4/ Without Brd7, cancer cells downregulate MHC-I, making them less visible to the immune system. At the same time, they secrete pro-metastatic cytokines, attracting inflammatory neutrophils and exhausted CD8+ T cells.
3/ In breast cancer metastasis models, Brd7 loss reactivates dormant metastatic cells in the lung, leading to aggressive tumor growth. But it’s not just about the tumor cells—the immune microenvironment also shifts dramatically.
2/ The study focuses on Brd7, a subunit of the PBAF chromatin remodeling complex. This complex regulates DNA accessibility, influencing how cancer cells behave in new tissues. But what happens when Brd7 is lost?
