Restoration of Capicua repressor activity or silencing of Capicua target genes ETV4 and ETV5 restores the sensitivity to trametinib. In addition, absence of Capicua creates new vulnerabilities that can be exploited to overcome resistance.
We show that absence of Capicua suppresses the requirement for Kras oncogene amplification during lung tumor development, leading to more efficient transformation of bronchiolar Club cells and resistance to MAPK pathway inhibitors.
