Stem-cell-derived beta cells mature metabolically upon murine engraftment - Diabetologia Aims/hypothesis The use of stem-cell-derived islets (SC-islets) as a source for cell-based therapy of type 1 diabetes shows great potential. However, SC-islets have a metabolically immature phenotype ...

News from our CoE by @otonkoskit.bsky.social lab, Eliisa VΓ€hΓ€kangas et al show that upon engraftment into mice stem cell derived pancreatic islets become insensitive to pyruvate, correlating with down regulation of the pyruvate/lactate transporter MCT1. link.springer.com/article/10.1... @helsinki.fi

Old mitochondria regulate niche renewal via Ξ±-ketoglutarate metabolism in stem cells - Nature Metabolism Andersson et al. show that intestinal stem cells enriched for old mitochondria are metabolically distinct and have enhanced ability to regenerate the epithelial niche.

News from our CoE by @katajistolab.bsky.social @simonsterson.bsky.social found that intestinal stem cells with old mitochondria make Paneth cells faster due to higher aKG levels and aKG improves regeneration in old animals by renewal of Paneth cells. www.nature.com/articles/s42... @helsinki.fi

Mitochondrial age-classes expand the heterogeneity of tissue-resident stem cells Nature Metabolism, Published online: 14 July 2025; doi:10.1038/s42255-025-01326-6During cell division, a subset of intestinal stem cells (ISCs) inherits old mitochondria that increase intracellular Ξ±-ketoglutarate levels. Ξ±-Ketoglutarate facilitates remodelling of DNA methylation and boosts the ability of ISCs to regenerate the stem cell niche. Metabolite intervention with Ξ±-ketoglutarate during ageing can replace defective niche cells and enhance regeneration.
