Additionally, as originally reported in our preprint, this study also characterized the signals elicited by LNPs in DCs that are important for the induction of Tfh cells as well as the type of DCs and the location of LNP uptake
Using an improved experimental approach, we also showed that the sensing of the mRNA shapes the quality of Tfh cell responses (heightened generation of IFN-Ξ³+ and IL-21+ Tfh cells) and favors more robust memory B cell and plasma cell responses, along with enhanced neutralizing antibody production.
In parallel, we found that, in a type I IFN-independent manner, signaling through IL-1R is also key for the induction of optimal GC responses to Spikevax, at least partially explaining our previous finding (www.cell.com/immunity/ful... that MyD88 is important in mRNA vaccination.
Indeed, none of the classical pattern recognition receptors (PRRs) that recognize nucleic acids in the endosome or in the cytosol are activated by Spikevax in vitro or required for GC responses in vivo. These data suggest the possibility that a less characterized PRR may sense the modified mRNA.
Our work also revealed that IRF3/IRF7 are necessary for the induction of type I IFNs and for GC responses to Spikevax, even though the upstream sensor recognizing the nucleoside-modified mRNA and leading to IFN production remains to be identified.
Among the new findings that emerged during the revision of this study is the observation that type I IFNs are crucial also for GC responses to Spikevax, one of the licensed mRNA vaccines for COVID-19.
In an earlier version of this work, also available at www.biorxiv.org/content/10.1..., we found that type I IFNs produced in response to the nucleoside-modified mRNA component act on DCs to modulate their maturation and the amplification of Tfh and GC B cell responses.
