Quantitative workflow to probe Bik1 conformational changes. Crosslinked (XL) peptides were obtained from Bik1 FL in dilute, phase separation, supernatant, and pellet conditions. Crosslinks specific to one condition (conformo-specific crosslinks) are then employed to monitor structural rearrangements in Bik1 samples using quantitative targeted proteomics
