This is great work. Really looking forward to how this evolves.

How does a silent genome know where to begin life?
A recent study by @evahoermanseder.bsky.social shows the early embryo is not a βblank slate.β H3K4me3 marks are pre-established in gametes and guide zygotic genome activation, acting as true epigenetic memory.

1/ π We got our Christmas present today: "Two distinct chromatin modules regulate proinflammatory gene expression" is now published @natcellbio.nature.com doi.org/10.1038/s415.... Our study introduces a scATAC-seq-based framework for genome-wide analysis of gene regulation features.
There's a super line up next March for the Keystone meeting in Geneva for anyone interested in chromatin and epigenetics! Not a bad spot for a Spring trip!

The Motamedi lab shows that Clr4 is recruited de novo to the ncRNA locus SPNCRNA230 even without Ago1 and pre-existing H3K9me. Sir2 and Clr3 (H3K9 HDACs) also accumulate there, and iterative cycles of H3K9 deacetylation then methylation spread a basal H3K9me state, which RNAi subsequently amplifies.
ππ£Join us for journal club tomorrow as Dr. Claudia Puri presents her work on "Transferrin receptor controls both autophagosome formation and closure via phosphatidylinositol 3-phosphate synthesis", a recent work published in Developmental Cell!

Fig. 1.Overview of TOR kinase signaling impacting chromatin. TOR phosphorylates a range of targets, including components of the translation machinery, downstream kinases, chromatin modifiers, transcription factors or metabolic proteins, thereby modulating their activity, localization, and nucleocytoplasmic shuttling. TOR signaling can also influence cytoskeletal dynamics, which in turn affects nuclear shape and structure. Colored elements indicate processes known to occur in plants, while those in gray refer to mammalian systems. Gray dashed arrows indicate uncertain or indirect pathways, while question marks indicate uncertain cellular localization of the process, highlighting potential areas for future research.

We show that JARID2 and PALI1 mimic H3K27me3 to antagonise PRC2 in vivo and restrict the spread of Polycomb domains.
