How does a silent genome know where to begin life?

A recent study by @evahoermanseder.bsky.social shows the early embryo is not a β€œblank slate.” H3K4me3 marks are pre-established in gametes and guide zygotic genome activation, acting as true epigenetic memory.

The Motamedi lab shows that Clr4 is recruited de novo to the ncRNA locus SPNCRNA230 even without Ago1 and pre-existing H3K9me. Sir2 and Clr3 (H3K9 HDACs) also accumulate there, and iterative cycles of H3K9 deacetylation then methylation spread a basal H3K9me state, which RNAi subsequently amplifies.
