Functionally treated CD54+ cells are able to engraft and contribute to muscle regeneration with a better efficiency compared to untreated cells.
We were able to purify SMPCs using a single surface marker (CD54/ICAM1) and analyze the cells transcriptionally and functionally.
At transcription level treated cells resemble late fetal stage SMPCs rather than embryonal SMPCs that are usually obtained in vitro
We started the project wondering if modulation of the jak2/stat3 pathway would be efficient in increasing the yield. When we inhibited the pathway after the appearance of PAX3+ cells (D12) we managed to double the amount of PAX7+ cells at D30, both in H9 and hiPSCs derived cultures
