Our study does not address the PL/LPS biosynthesis/transport coordination. However, talking about coordination, we speculate that the lipid tails of LptM help prevent that PLs of the OM inner leaflet access the lateral gate of LptD when this opens to release LPS.

The small lipoprotein LptM intercalates the LptD lateral gate promoting gate dynamics and access by LPS. LptY (YedD) stabilizes the beta-taco domain and contributes together with LptM to optimal functioning of the Lpt transenvelope bridge.

The LptD beta-strand 1, defining the mobile side of the lateral gate, binds LPS and performs a stroke movement toward the external leaflet during the contracted-to-extended state transition.

In this paper we show that LptD functions via a conformational switch of the beta-taco/beta-barrel interface, which alternates between contracted and extended states.
