If you're studying #RNA transcript isoforms, check out IsoVis (the Isoform Visualiser webserver). isomix.org/isovis/

Recent updates now also allow visualisation of:
1. RNA modification sites and levels
2. Peptides mapping to open reading frames in isoforms and their abundances. πŸ§ͺ

No prob! Definitely doable, dRNA will be less biased than cDNA but may not get you the best depth. We had a post-mortem brain sample at RIN 7.8 and it still looked ok. I'd just advise using the RIN as a cofactor in analysis if you have varied samples.

Pervasive effects of RNA degradation on Nanopore direct RNA sequencing Abstract. Oxford Nanopore direct RNA sequencing (DRS) is capable of sequencing complete RNA molecules and accurately measuring gene and isoform expression.

You can check out our paper here but we only went to a minimum of 7: doi.org/10.1093/narg.... Probably wouldn't advise doing it with much lower... depends what you want to see!

Spatial transcriptomics and genetically implicated genes identify putative causal tissue structures for complex traits Spatially resolved transcriptomics is transforming our understanding of cellular and molecular diversity of tissues. Here, to identify tissue structures that are enriched for putatively causal disease...

New preprint! My (now former) postdoc @kvastad.bsky.social led this integration of GWAS and spatial transcriptomics (ST) data to identify tissue structures with enrichment of disease-implicated genes = likely causal drivers of disease biology.
