I recently reviewed a paper where the corresponding author was SO blunt, bordering rude, & started every sentence like they hated me instead of "thank you my king for the comment", and I was like FAIR, I'm here to comment on the science, and don't have a micropenis ego that can't handle this tone.

Thanks a lot for sharing your experiences, Stefan! That is really weird with H3K27ac. That's the very modification I would have considered a no-brainer in no matter what assay. What are the issues? Fewer peaks, peak height, noise...all combined?

1. Meticulously annotating cell clusters
2. Extracting regulatory programs in terms of coordinated gene sets and accessible regulatory elements
3. Using deep learning to identify regulatory syntax
4. Resolving context-specific TF activity
