Levels of lysine malonylation (MaK) and its regulator Sirt5 were examined in mouse and in vitro cartilage samples. Results showed ↓SIRT5 and ↑MaK in cartilage during aging. In mice, combination of Sirt5 deficiency & obesity exacerbated joint degeneration

My friend Mick Jurynec at Utah also independently identified a human Sirt5 coding mutation that is linked to familial OA. We will follow up on this work using a mouse model carrying this mutant allele to further study its functions.
We used a combination of systemic and cartilage conditional KO mouse models to demonstrate that Sirt5-MaK is an important regulator for chondrocyte metabolism and OA development during obesity.
