Inhibition is one of the main confounding factors in eDNA work due to increased risk of false negatives (Type II Error). I found diluting samples does not work well as it over-dilutes the samples. We've been using Recombinant Albumim (RA) as of late. #eDNA
I can now add "extracted DNA from dog food" to my CV. #eDNA

I'm with the Marine Gen Probe Lab at Dalhousie University in Nova Scotia, Canada and we do a lot of #eDNA detection work in freshwater lakes around the province, especially within the Petite Rivière. My job is processing samples and optimizing methods.

When you have three multiplex PCRs to do, each one takes three hours, and your third machine decides to stop working so you spend 3 hours longer than planned on your work.
My day yesterday.
Note: we have a Main Lab space with our gDNA samples and a Clean Lab space with no gDNA (minus qPCR positives kept under quarantine). The Main Lab is where the contamination risk is.
For a previous experiment we put out several of these devices around our laboratory space to determine if we could gather detectable levels of fish DNA from the air (i.e. is aerial contamination of eDNA samples a risk).
The answer was yes for at least Atlantic Salmon and Atlantic Whitefish.
