@hakha.bsky.social and I wrote a Research Briefing (with a lay summary + "behind the scenes") of our paper on how genes are prioritized by GWAS and rare variant burden tests. ๐งฌ๐งช

Brilliant paper by Visscher et al.
Populations differ in traits/disease burden. Are these differences due to genetics?
Comparing single variants or polygenic scores between populations is biased due to environmental confounders correlated with the variants.

A few thoughts on Herasight, the new embryo selection company. First, their whitepaper (drive.google.com/file/d/1EpFi...) implies that competitors like Nucleus have been marketing and selling grossly erroneous risk estimates. This is shocking if true! ๐งต
What's going on in psychiatric genetics? The @pgcgenetics.bsky.social have now published our latest progress review in @thelancetpsych.bsky.social, highlighting discovery, and laying out a map for moving forwards!

You donโt need more ideas.
You need a better question.
A thread on clarity, chaos, & what a Nobel Prize winner, a mosquito net & a PhD meltdown taught me about writing (+ thinking) better.

We hope it can be used in other conditions to identify the best timing for intervention.๐ง

This is the first application of network temperature ๐ก๏ธ in mental health and provides a network-wide alternative to node-focussed measures (centrality estimates) which lack applicability in mental health conditions...
Our findings highlight the criticality of early intervention in young people when symptoms are more flexible and responsive to intervention or treatment ๐ฉบ
Depression is not just about the type of treatment - but the timing! โณ...
In 3 large longitudinal adolescent cohorts (@CO90s, @theabcdstudy MCS @CLScohorts) we found that temperature decreased across adolescence, meaning symptoms became more aligned and stable with age ๐๐ง ...
We applied this to depression symptoms:
In a high temperature network, symptoms are less stable and fluctuate ๐ฅ
In a low temperature network, symptoms are stable and harder to shift ๐ง ...
Think of the transition from solid>liquid>gas: As temperature increases, particles become more random and less stable ๐ฅต. As temperature decreases, the system becomes more rigid and fixed ๐ฅถ...
Depression is dynamic - symptoms interact over time. We can model depression using networks which capture symptom-symptom relationships. ๐ธ๏ธ
We applied a concept from statistical thermodynamics - temperature ๐ก๏ธ ...
Excited to share that the second paper from my PhD has just been published in @NatMentHealth!๐ง ๐งต
We modelled depression symptom networks in >35k teenagers to derive their 'network temperature' and found evidence symptom stabilisation across adolescence...

You've heard of DNA, but what about GNA? Very excited about this preprint with @jgthorp.bsky.social and others that introduces Genomic Network Analysis (GNA), an open-source multivariate tool for performing network analysis using GWAS summary statistics as input. 1/2

Hello from Singapore #wcpg2024!๐ I am presenting an update on the current results of the *Adolescent-Onset Depression GWAS* on Friday at 3pm in the symposium on Genomics and Life Course Data. Come and watch!
Iโm presenting three different projects at these upcoming conferences over the next month! Looking forward to meeting fellow MH researchers ๐

They made a nice recording & allowed me to share it.

Great - thank you sm! I have performed MI and taken the mode of the variable combinations as final dataset >> Ising. Hopefully arguable given nature of the data/model? Then will run sensitivity with complete cases, with and without max likelihood estimator. Will look at IRT approaches too ๐
@eikofried.bsky.social any advice? ๐๐
Missing data, longitudinal binary symptoms for multi-group Ising model estimation. HELP!!!! Impute? If so, how to pool estimates? Don't think it's right to avg. over networks? Use maximum likelihood estimator instead? ๐ตโ๐ซ
We are also incredibly grateful to the participants and staff of the #ALSPAC and #ABCDstudy, to the PGC, 23andMe, the funders MRC and the reviewers! ๐
