Here we show that reduced 4EBP1 leads to acquired Capivarsertib resistance via increased cap-dependent protein synthesis.

We also developed Single Molecule Analysis of ssDNA gaps (SMAss) that quantitatively resolved ssDNA gap dynamics.

We demonstrate that the 9-1-1 complex safeguards BRCA2-deficient cells from lethal ssDNA gap accumulation by suppressing PRIMPOL-dependent gap formation, restraining EXO1 degradation and promoting POLζ-mediated filling.
