Combinatorial BMP4 and activin direct the choice between alternate routes to endoderm in a stem cell model of human gastrulation Inge et al. show that human endoderm originates from two converging developmental routes with distinct dynamics and efficiencies yet similar developmental potential. Combinatorial activin and BMP4 sig...

1/7 Really happy to see my PhD work published in ✨Developmental Cell✨ today! We find human endoderm is specified by two developmental trajectories 🔀, and the choice between alternate routes is dictated by the combinatorial BMP4/Activin signalling. www.cell.com/developmenta...

Transgene-free generation of mouse post-gastrulation whole embryo models solely from naive ESCs and iPSCs Hanna and colleagues show that signaling pathway modulation amplifies the inherent flexibility of genetically unmodified mouse naive PSCs to adopt extra-embryonic cell fate potential. This endows them...

1/n Fun journey continues: Our new @cp-cellstemcell.bsky.social paper out on how the Inherent & Dormant Plasticity of Mouse Naive PSCs can be "Exaggerated" to Directly form ~E8.5 Transgene-Free (TF) Mouse SEMs & WITHOUT inducing 8-16-Morula-like "Embryo Founder Cells".

So happy to finally see this work published! We reveal that an in vitro system mimics specification of extra-embryonic mesoderm into the visceral yolk-sac and allantois, and use this model alongside in vivo experiments to study the role of Eomes and T in these tissues. Big thanks to all co-authors 🥳

Programmatic design and editing of cis-regulatory elements The development of modern genome editing tools has enabled researchers to make such edits with high precision but has left unsolved the problem of designing these edits. As a solution, we propose Ledi...

Our preprint on designing and editing cis-regulatory elements using Ledidi is out! Ledidi turns *any* ML model (or set of models) into a designer of edits to DNA sequences that induce desired characteristics.

Dissecting regulatory syntax in human development with scalable multiomics and deep learning Transcription factors (TFs) establish cell identity during development by binding regulatory DNA in a sequence-specific manner, often promoting local chromatin accessibility, and regulating gene expre...

Delighted to share our latest work deciphering the landscape of chromatin accessibility and modeling the DNA sequence syntax rules underlying gene regulation during human fetal development! www.biorxiv.org/content/10.1... Read on for more: 🧵 1/16 #GeneReg 🧬🖥️

Huge congrats to Anusri! This was quite a slog (for both of us) but we r very proud of this one! It is a long read but worth it IMHO. Methods r in the supp. materials. Bluetorial coming soon below 1/
