We just wrapped up the 7th Molecular & Cellular Pharmacology Retreat in the Florida Keys, and the energy was incredible.
Under the theme “Colab in the Cays,” scientists from 15+ labs shared 53 talks across two days, with every single participant presenting their work.

These mutations mimic protonation (i.e., activation) and, along with abolishing Na+ negative allosteric modulation, were intended to fully activate GPR4, 65, and 68 precisely as they did
There is a misunderstanding here. These mutations were not designed to abolish proton-mediated activation but to achieve the exact opposite based on the predicted physical chemistry and self-energy considerations.
"A recent study employing parallel mutagenesis of titratable residues in GPR68 identified a conserved triad of buried acidic residues in proton sensing GPCRs. Here, too, neutral mutations to these sites shift pH50 but do not abolish proton-mediated receptor activation."
