Membrane editing with proximity labeling reveals regulators of lipid homeostasis - Nature Chemical Biology Coupling an optogenetic lipid-modifying enzyme with proximity labeling reveals protein networks and mechanisms regulating lipid homeostasis in the membranes of target organelles.

Thrilled to share our latest study, led by @reikatei.bsky.social, in @natchembio.nature.com! We began by asking a simple questionβ€”how do cells know if they have too much of a lipid in a particular membrane, and how do they respond to rectify this imbalance?

Our latest story: a protein-protein interaction between an E3 ubiquitin ligase and an autophagy adaptor affects liquid-liquid phase separation and sequestration of a key mediator of antioxidant signaling β€” but not via ubiquitination! Bravo to Lin Luan and Xiaofu Cao!

My personal #lipidtime highlight is antigen-gated control of Gq/DAG signaling!🚦 Using PAGER-Gq, we showed DAG production driven by soluble and surface antigens in HEK cells and neurons. Plus, neuron images reveal local DAG production at antigen-contact membranes! Can it be something useful?πŸ€”

Our paper about the development of programmable antigen-gated G-protein-coupled engineered receptors (PAGERs) is published today! www.nature.com/articles/s41...
PAGERs couple detection of a wide range of custom-defined antigen to a variety of output including transgene expression and cell signaling.

Could one envision a synthetic receptor technology that is fully programmable, able to detect diverse extracellular antigens – both soluble and cell-attached – and convert that recognition into a wide range of intracellular responses, from gene expression and real-time fluorescence to modulation..
