A longitudinal single-cell atlas to predict outcome and toxicity after BCMA-directed CAR T cell therapy in multiple myeloma Rade et al. present a longitudinal single-cell atlas of BCMA CAR-T therapy in multiple myeloma, revealing that cilta-cel drives potent CD4⁺ cytotoxic responses and superior outcomes. Their findings identify plasmacytoid dendritic cells as unexpected BCMA-expressing targets and suggest therapeutic potential in blastic plasmacytoid dendritic cell neoplasm.

Macrophage metabolic exhaustion and PANoptotic cell death drive chronic tissue inflammation in rheumatoid arthritis Rheumatoid joints contain macrophages with reparative potential, yet inflammation persists. Huang et al. demonstrate that these homeostatic macrophages lose their protective role by undergoing PANoptotic death. Mechanistically, C1q activates the mitochondrial enzyme SARM1, driving NAD⁺ cleavage into cADPR, which precipitates metabolic collapse and PANoptosis. The resulting macrophage death amplifies autoimmune tissue inflammation through the release of pro-inflammatory mediators.

Ubiquitination-directed cytosolic DNA degradation governs cGAS-STING-mediated immune response to DNA damage Li et al. show that SPOP mutations and USP7 overexpression promote TREX1 upregulation, leading to increased cytosolic DNA degradation and suppression of cGAS-STING-mediated immune activation. Targeting USP7 enhances the effectiveness of radiotherapy combined with immune checkpoint blockade, providing a new strategy to overcome resistance and improve therapy outcomes.

It all started with an unexpected observation 🔍:
