Be intellectually humble — if you are studying an interesting question, remember that the question is bigger than one’s intellect.

Are authors fairly judged by assessing the #journals in which their work is published? @bihutchins.bsky.social &co reveal that most influential papers are published in lower tier journals, and more authors would be better recognized with #ArticleLevelMetrics #ALMs @plosbiology.org 🧪 plos.io/4oV58Ed

Left: Graphic summary of the USPPAR approach. The entire process is divided into three stages. Dissociation involves separating whole cells from culture dishes or extracting nuclei from tissues, followed by fixation. Barcoding includes converting mRNA to cDNA with initial tagging, then performing 2–3 additional rounds of barcoding through ligation. Amplification encompasses cell/nucleus lysis, high-efficiency poly(dA) extension of cDNA, preamplification, tagmentation, and final library amplification. Right: Joint UMAPs of 50,584 murine hepatic nuclei, including 12,646 nuclei from each technology: USPPAR_pH3 (pH3 lysis), USPPAR_CuC (CuC lysis), Liver Cell Atlas (LCA, 10× Chromium), and PanSci (EasySci platform). Nuclei are colored by technology (Technology)

Single-cell RNA-sequencing based on combinatorial barcoding is pricey and lacks sensitivity. USPPAR is a novel #scRNAseq approach that combines split-pool #barcoding & optimized polydeoxyadenylation, outperforming existing methods without specialized equipment @plosbiology.org 🧪 plos.io/3XUsleH
