Rewiring natural killer cells for next-generation cancer therapies Harnessing the cytotoxic potential of natural killer (NK) cells for cancer immunotherapy has proven challenging. In this issue of Cancer Cell, Biederstädt et al. and Nikolic et al. utilize genome-wide CRISPR screening to uncover novel regulators of NK cell function, paving the way for developing next-generation NK cell therapies.

Small-molecule RNA therapeutics to target prostate cancer Kuzuoglu-Ozturk et al. showcase a therapeutic approach, “translatome therapy,” to combat lethal prostate cancer. Zotatifin, a clinical molecule, targets the RNA-helicase eIF4A. Zotatifin decreases the select translation of key oncogenes, androgen receptor (AR), and HIF1α through remodeling their 5′ UTR RNA structure. Zotatifin enhances anti-androgen and radiotherapy sensitivity, highlighting a promising anti-cancer treatment.

Translation dysregulation in cancer as a source for targetable antigens Weller et al. substantiate the role of translation dysregulation in mediating anti-tumor immunity by demonstrating that TYW2 loss in melanoma cells induces aberrant peptide MHC presentation, increasing both tumor immunogenicity and sensitivity to ICB therapy. TYW2 expression levels in patients with melanoma predict clinical outcomes, highlighting translation regulatory factors as immunotherapy targets.

Translation dysregulation in cancer as a source for targetable antigens

Spatial and multiomics analysis of human and mouse lung adenocarcinoma precursors reveals TIM-3 as a putative target for precancer interception Zhu et al. reveal coordinated interplay between innate and adaptive immunity in the transition from precancer to invasive lung cancer. TIM-3 upregulation peaks at precancer stage and its blockade enhances anti-tumor immunity while inhibiting lung precancer progression, suggesting TIM-3 as a potential target for lung precancer interception.
