Our findings highlight that PDAC cells exchange components of the translation machinery and mRNA via tunnelling nanotubes, partially restoring protein synthesis in translation-impaired cells and revealing TNT dynamics as a potential therapeutic target in pancreatic ductal adenocarcinoma. 💜

In this study, we show that pancreatic cancer cells can transfer polyadenylated mRNA, 5.8S rRNA, ribosomal proteins and assembled 80S ribosomes via tunnelling nanotubes, thin membranous intercellular channels that can be induced by sub-IC50 doses of gemcitabine.

We are looking for highly motivated Ph.D. candidates
We have an open PhD position in my lab to work on pancreatic cancer biology, specifically metabolic cooperation via tunnelling nanotubes and the development of resistance to treatment. For more info follow the link below.

Ribosome transfer via tunnelling nanotubes rescues protein synthesis in pancreatic cancer cells
Background: Pancreatic ductal adenocarcinoma (PDAC) is considered as one of the deadliest types of cancer. Tunnelling nanotubes (TNTs) are thin, membranous, intercellular communication structures obse...
Our significantly expanded preprint is out. In it we show that pancreatic cancer cells can transfer ribosomes via tunnelling nanotubes and rescue protein synthesis when co-cultured with cells with impaired ribosomal biogenesis.

I'm thrilled to share that our story is now out in Science Advances! 🎉 We use quantitative imaging to map the mito central dogma, define translation hubs in the mitochondrial matrix, and show that they're replaced by Mitochondrial Stress Bodies (MSB) when mtRNA processing is perturbed 1/4

Screenshot Ron ThermoFisher page on CellTracer dyes saying there is no transfer to unstained cells.
Fluorescence microscopy picture of mixed stained/unstained cells with CellTracer Green showing partially stained cells due to intercellular transfer of the dye.
ThermoFisher tell us their CellTracker dyes are not transferred to neighbouring cells but we see significant transfer @24h when we mix stained with unstained cells. Any suggestions how to label cells reliably so that we can track distinct populations with no or minimal transfer? 🧪 #fluorescence

Long-term imaging of individual ribosomes reveals ribosome cooperativity in mRNA translation
Ribosomes cooperate through transient collisions to ensure efficient translation.
Our paper on Stopless-ORF Circular RNAs (socRNAs) is now out in Cell. By high-res tracking and comparing translation by either single or multiple ribosomes, we find that ribosomes cooperate to overcome pausing to ensure fast and efficient translation

A new mechanism for cancer-induced immune evasion. We knew about cancer cells hijacking mitochondria. Now we learn about how their mitochondrial transfer to T cells impairs the tumor immune response.

Instead of listing my publications, as the year draws to an end, I want to shine the spotlight on the commonplace assumption that productivity must always increase. Good research is disruptive and thinking time is central to high quality scholarship and necessary for disruptive research.

A quick snapshot from a fluorescence microscope, taken during a teaching session with medical students, showcased the fascinating world of mitochondria. The students were excited, asking many questions about why cells transfer mitochondria (MitoTracker-Red) via tunneling nanotubes (actin-green).

#TunnelingNanotubes are long filamentous actin-based cell protrusions that create communication networks between cells, especially in cancer. Here, an especially long nanotube transmits GFP-expressing oncolytic virus, a mechanism for propagation of the bystander effect.
