At this high resolution, we also caught something new: Using the new #HAK-Actin #UExM probe, we saw that
#Actin accumulates at the furrow base before compartments fully seal ➡️ they're still open at the bottom.

➡️ The #multicellular epithelium-like stage is shorter than we previously proposed.

Using #UExM, we could finally see the MT network. Two populations:
Cortical MTs=from MTOCs to the cortex, spacing nuclei evenly
Furrow-tracking MTs=elongating with the ingressing membrane.

We recently investigated how much and when tintinnid ciliates produce shell material during the cell cycle to construct a new shell (lorica) after division: tinyurl.com/49usp8xw
We adapted a classical staining technique.

Its not the easiest sample, some protective layer hinders homogenous ExM of the cell body all the time,but the reaction we got when it worked was worth it n/2
