Something I am always baffled by: Center for Biologics Evaluation and Research (CBER) in the FDA does *NOT* regulate antibody or other protein therapies.
That's done by Center for Drug Evaluation and Research (CDER) - even though they're biologics.

They have created: PK/PD, PBPK, PBAM, PBBM, QSP, QST...all of which are related but different concepts or frameworks.

We talk a lot about the reasons drug candidates fail, but I feel like we don't talk enough about WHY specific candidates fail.
Things like "lack of efficacy" or "tox" aren't really actionable. Was it binding affinity? Binding to a homolog? Metabolized too fast? A mix?
