Clinical data from lung adenocarcinoma patients with KRASG12C and #STK11 #LKB1 mutations were analyzed, looking for correlation between high squamous gene signature at baseline and response to adagrasib. Analysis reveals patients with a high squamous gene signature had poorer responses to treatment.

Targeting mTOR and Glycolysis in HER2-Positive Breast Cancer
If LKB1 is removed (or "knocked out"), the activation of #AMPK by PKA is significantly reduced. However, even without #LKB1 ( #STK11 ), there is still some activation of AMPK.
The research found that high expression levels of certain genes associated with #LKB1 signaling, such as #AMPK, #LYK5, and #PAK1, correlated with improved survival rates across all breast cancer types studied.
Different breast cancer subtypes respond differently to #LKB1 signaling. For instance, while some genes like #PAK1 and #SIK2 are linked to improved survival in Triple-Negative Breast Cancer (TNBC) before chemotherapy, their impact changes after treatment.
Authors evaluated #LKB1 signaling pathway gene expression associations with patient survival in overall breast cancer, specific subtypes, as well as pre- and post-chemotherapy.
Understanding the metabolic consequences of #LKB1 inactivation could provide critical insights for developing targeted therapies in this #cancer type
Given that current immune checkpoint inhibitors (ICIs) are less effective in patients with #LKB1 and #KEAP1 mutations, there is a pressing need for new treatment approaches.
