Structural Changes in Blood Proteins Could Enable Early Detection of Alzheimer’s Disease Researchers at Scripps Research Institute have developed a novel blood test that may detect Alzheimer’s disease earlier than current methods by analyzing subtle structural changes in proteins rather than their levels. The study, involving over 500 participants, found that three proteins in the blood—C1QA, clusterin, and apolipoprotein B—showed structural changes that strongly correlate with Alzheimer’s progression. Using mass spectrometry and machine learning, the team was able to differentiate cognitively normal individuals from those with mild cognitive impairment or Alzheimer’s with high accuracy, exceeding 93% in some comparisons. These findings suggest that monitoring protein folding disruptions in the blood could serve as a predictive biomarker, complementing traditional amyloid beta and tau tests. Repeat tests confirmed the reliability of this protein structural score over time and its correlation with cognitive performance and, to a lesser degree, brain shrinkage seen on MRI scans. While the research is promising, larger studies with longer follow-up periods are necessary before clinical application. The approach could eventually help diagnose Alzheimer’s at an earlier stage, track disease progression, and evaluate treatment effectiveness. Researchers are also exploring whether similar structural profiling could apply to other diseases such as Parkinson’s and cancer.
